Coligh’s fluorescence microscopy filters

Coligh's fluorescence filters are a collection of high-performance filter products that have been specifically designed to work with a wide range of popular fluorophores, as well as some of the most promising new ones. These filters have been used as the starting point for creating custom-designed filters for some of the most advanced instruments on the market, including DNA sequencers, cytometers, microplate readers, in vivo imagers, and thermocyclers.

At Coligh, we are now offering our OEM-proven designs to individual researchers through our catalog of high-performance filter sets for fluorescence microscopy. Our filters are manufactured using hard coating technology, which provides excellent spectral performance, spectral stability, and physical durability.

Our filters are designed with several key features that ensure maximum performance. First and foremost, we prioritize exceptionally high passband transmission to ensure that as much of the desired signal as possible is detected. We also use maximized out-of-band blocking, including extended range blocking on excitation filters, to minimize background noise and signal interference. Additionally, we employ extremely steep slopes from rejection to transmission regions, which enables the filter to sharply and accurately discriminate between desired and undesired signals.

We also focus on optical finishing to achieve maximum image rendition, including precision grinding and polishing, and anti-reflection coatings to reduce the amount of light lost due to reflection. Our filters have specific bandpass specifications of center wavelength and bandwidth, which provide maximum image brightness and minimum background noise.

Fluorescence Filter Sets

A typical fluorescence filter set consists of an excitation filter, an emission filter, and a dichroic filter. Here's a brief description of each:

Excitation Filter: This filter is placed in the light path to select a specific wavelength range of light for excitation of the fluorophore. Excitation filters typically have a narrow bandwidth and high transmission at the desired excitation wavelength, while blocking other wavelengths of light that could potentially excite the fluorophore.

Emission Filter: This filter is placed in the light path to select a specific wavelength range of light emitted by the fluorophore. Emission filters typically have a narrow bandwidth and high transmission at the desired emission wavelength, while blocking other wavelengths of light that could potentially interfere with detection of the emitted light.

Dichroic Filter: This filter is placed at a 45-degree angle to the light path to separate the excitation and emission light. It reflects the shorter wavelength excitation light towards the sample while allowing the longer wavelength emission light to pass through to the detector.

When selecting fluorescence filters, it is important to consider factors such as the excitation and emission spectra of the fluorophore, the spectral properties of the light source and detector, and any potential spectral overlap between different fluorophores in a multi-color imaging experiment.
